Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
This supplement contains one active ingredient: pregnenolone 50 mg per capsule. Pregnenolone is a hormone precursor — your body naturally produces it and uses it to make other hormones like estrogen, progesterone, and testosterone.
The capsule itself is made from gelatin and filled with microcrystalline cellulose, vegetable stearate, and silica as inactive ingredients.
Does it work?
Couldn't assess
The evidence we hold shows insufficient reliable data to rate pregnenolone for any of the conditions it's marketed for — including Alzheimer disease, back pain, bipolar disorder, autism spectrum disorder, and alcohol use disorder. That doesn't mean it doesn't work; it means we don't yet have strong clinical evidence showing whether it does.
How safe is it?
Well-documented data
Pregnenolone is generally well tolerated when taken by mouth. The most common side effects reported in users include acne, agitation, diarrhea, drowsiness, excitement, hair loss, sweating, and tremor.
A few cases of skin rash with a burning feeling have been reported, possibly related to sun sensitivity — these cleared on their own. Because pregnenolone is a hormone with limited human safety data, use only under guidance from a healthcare professional.
The safety data advises against taking it during pregnancy due to unknown risks. There isn't enough information to know if it's safe while breastfeeding — talk with your doctor or pharmacist about whether it's right for you in that situation.
Meds to double-check
Moderate interaction found
Before taking this product, check with your doctor or pharmacist if you take estrogens, progestin, progesterone, testosterone, or benzodiazepines (sedatives). Pregnenolone may enhance the effects of hormone medications and may weaken how well benzodiazepines work.
The bottom line
Scorecard at a glanceFully disclosed formula with no assessable stated purpose. Moderate medication interactions have been identified, and safety information is well characterized.
Pregnenolone might appeal to someone looking to support hormone balance or address one of several conditions, though evidence for those uses isn't established yet. If you take any estrogen, progestin, progesterone, testosterone, or benzodiazepine medications, this is not a product to start without checking first.
Talk with your doctor or pharmacist about whether pregnenolone makes sense for you and your current medications.

Pregnenolone 50 mg by Life Extension Drug Interactions
HelloPharmacist Interaction Report
Life Extension Pregnenolone 50 mg interacts with several hormone-based medications through its pregnenolone content.
The most serious concern is a moderate interaction with estrogens — pregnenolone is a precursor to estrogen, so taking it may enhance estrogen's effects in your body, which could amplify both therapeutic and unwanted effects.
Read the full breakdown — every affected drug type, severity by severity
Pregnelolone also has moderate interactions with progestin, progesterone, and testosterone. Like estrogens, these are hormones your body can make from pregnenolone, so the supplement might strengthen their effects.
There's one additional moderate interaction to know about: benzodiazepines (sedatives like diazepam). Early research suggests that regular pregnenolone use may actually weaken how well these drugs work, possibly by changing how your brain's GABA receptors respond.
Altogether, these interactions span 82 individual medications. Before you start this supplement, run your exact hormone medications and any benzodiazepines through the interaction checker below.

For each ingredient in Pregnenolone 50 mg with known interactions, here are the types of medications it can affect. Open any type for the detail — or search your exact drug in the checker above.
Pregnenolone
Benzodiazepines
Concomitant use of pregnenolone may reduce the effects of benzodiazepines.
Very preliminary clinical research shows that chronic use of pregnenolone reduces sedative effects of diazepam when compared with chronic use of placebo. This effect may be related to the activity of pregnenolone at GABAA receptors.
Estrogens
Theoretically, taking pregnenolone might enhance the effects of estrogens.
Pregnenolone is a precursor for several steroid hormones, including estrogens.
Progesterone
Theoretically, taking pregnenolone might enhance the effects of progesterone.
In humans, some research shows that oral administration of pregnenolone can increase levels of progesterone. However, other research shows no effect on progesterone levels. It is possible the impact of pregnenolone on progesterone levels may be dose dependent.
Progestin
Theoretically, taking pregnenolone might enhance the effects of progestin.
Pregnenolone is a precursor for several steroid hormones, including progestin.
Testosterone
Theoretically, taking pregnenolone might enhance the effects of testosterone.
Pregnenolone is a precursor for several steroid hormones, including testosterone. However, preliminary clinical research shows that taking pregnenolone 30-500 mg orally daily for up to 8 weeks does not affect testosterone levels.
