Biotin is generally well tolerated, even at higher doses, because your body excretes excess in the urine. High-dose biotin has rarely caused mild diarrhea; one case report linked a combination of biotin and pantothenic acid to a serious inflammation (eosinophilic pleuropericarditis), but it's unclear whether biotin was the culprit.

Taurine is generally well tolerated short-term in healthy adults, though long-term safety data is less certain. Common side effects include constipation, diarrhea, and indigestion.

Rare case reports describe allergic reactions in people with sulfa or sulfite sensitivities. R-lipoic acid is generally well tolerated but commonly causes headache, heartburn, nausea, and vomiting.

Rare case reports raised concerns about an immune reaction to insulin (insulin autoimmune syndrome), and topical use can cause rash or skin irritation. For pregnancy, biotin is likely safe in normal dietary amounts, and taurine is likely safe, but R-lipoic acid has insufficient safety data — the safety notes advise against use in pregnancy and breastfeeding.

Alkylating Agents

Theoretically, the antioxidant effects of alpha-lipoic acid might alter the effectiveness of alkylating agents.
The use of antioxidants like alpha-lipoic acid during chemotherapy is controversial. There are concerns that antioxidants could reduce the activity of chemotherapy drugs that generate free radicals. However, some researchers theorize that antioxidants might make chemotherapy more effective by reducing oxidative stress that might interfere with apoptosis (cell death) of cancer cells. More evidence is needed to determine what effect, if any, antioxidants such as alpha-lipoic acid have on chemotherapy. Advise patients to consult their oncologist before using alpha-lipoic acid.
