Encorafenib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Cannabinol (cbn) interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of cannabinol.
In vitro research shows that CYP3A4 enzymes are involved in the 8-hydroxylation of cannabinol. So far, this interaction has not been reported in humans.

Here's the key thing to keep straight: in this interaction, it's the encorafenib (Braftovi) side that may affect the CBN, not the other way around. Encorafenib can act as a CYP3A4 inhibitor, and lab studies suggest CYP3A4 helps break down cannabinol. So in theory, encorafenib could slow CBN's clearance and let it build up, which might mean more drowsiness, grogginess, or dizziness from the CBN.

I'll be honest with you, though: this is only based on test-tube research and has never actually been reported in people, so the real-world risk is uncertain. My bigger concern is that encorafenib is a serious cancer medicine with its own important interactions. Please loop in your oncology pharmacist or doctor before adding CBN.

About Encorafenib

What Is Encorafenib Used For?

Brand name: Braftovi

Braftovi is used to treat cancers that have a specific change (mutation) in a gene called BRAF — specifically the V600E or V600K mutation, which must be confirmed by an FDA-authorized test before starting treatment. It is always used in combination with other medicines, never alone as standard therapy.

Braftovi (in combination with binimetinib) treats unresectable or metastatic melanoma with a BRAF V600E or V600K mutation. For metastatic colorectal cancer (mCRC) with a BRAF V600E mutation, Braftovi is used either with cetuximab and fluorouracil-based chemotherapy, or with cetuximab alone after prior therapy. Braftovi (in combination with binimetinib) also treats metastatic non-small cell lung cancer (NSCLC) with a BRAF V600E mutation.
