Lonafarnib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Cannabinol (cbn) interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of cannabinol.
In vitro research shows that CYP3A4 enzymes are involved in the 8-hydroxylation of cannabinol. So far, this interaction has not been reported in humans.

Here's the key thing to understand: this interaction is about how Lonafarnib (Zokinvy) could affect the supplement, not the other way around. Lonafarnib blocks an enzyme called CYP3A4, and lab studies suggest that same enzyme helps your body break down CBN. So in theory, taking them together could let CBN build up a bit, which might make you feel more of its effects, like extra drowsiness or grogginess.

I want to be honest about how thin the evidence is here. This comes only from test-tube research, and it has never actually been reported in people. Still, since Lonafarnib is a serious medication with its own important rules, I'd run any CBN product past your pharmacist or doctor first before combining them.

Theoretically, cannabinol might increase the levels and clinical effects of CYP2C9 substrates.
In vitro research shows that cannabinol potently inhibits CYP2C9 enzymes. So far, this interaction has not been reported in humans.

Here's the simple version. Lonafarnib (Zokinvy) is cleared from your body partly by a liver enzyme called CYP2C9. Lab studies suggest CBN can block that same enzyme. If it does that in people, your body would clear lonafarnib more slowly, so the drug could build up and its side effects could get stronger. Think more nausea, vomiting, diarrhea, poor appetite, and tiredness.

I want to be honest about how strong this is: it's theoretical, based only on test-tube work, and it hasn't been seen in real patients yet. Still, lonafarnib is a serious medication, so don't add CBN on your own. Run it by your doctor or pharmacist first.

About Lonafarnib

What Is Lonafarnib Used For?

Brand name: Zokinvy

Zokinvy (lonafarnib) is approved for patients 12 months of age and older who have a body surface area of 0.39 m² or above. It is used to reduce the risk of mortality in Hutchinson-Gilford Progeria Syndrome (HGPS) — a rare, severe premature-aging disorder.

Zokinvy is also used to treat processing-deficient Progeroid Laminopathies — a related group of rare genetic conditions — specifically those caused by a heterozygous LMNA mutation with progerin-like protein buildup, or by homozygous or compound heterozygous ZMPSTE24 mutations. It is not indicated for other Progeroid Syndromes or processing-proficient Progeroid Laminopathies.
