Here's the basic idea with European mandrake and entrectinib (Rozlytrek). Mandrake's tropane alkaloids (atropine-like compounds) slow down how fast your gut moves. When food and pills sit in your digestive tract longer, your body may have more time to soak up a drug. So in theory, mandrake could let you absorb a bit more entrectinib than usual, nudging its levels up.

Entrectinib is active on its own, not a prodrug, so more absorption could mean stronger effects and more side effects like dizziness, tiredness, an irregular or fast heartbeat, and changes on a heart tracing (QT prolongation). I'll be honest though, this is a theoretical concern based on how these compounds behave, not on real-world reports. And separately, mandrake itself is genuinely toxic, so please don't take it. Talk with your oncology team before adding anything.

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC) — a type of lung cancer where the ROS1 gene is rearranged — as confirmed by an FDA-approved test.

Rozlytrek is also approved for adult and pediatric patients older than 1 month with solid tumors that have an NTRK gene fusion (a specific genetic alteration), detected by an FDA-approved test, that have spread or cannot be removed without causing serious harm, and have worsened after prior treatment or have no good treatment alternatives. This NTRK indication was granted accelerated approval based on tumor response rates; continued approval may depend on results from confirmatory trials.
