Pitolisant Hydrochloride falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Evodia interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of evodia.
Animal research shows that concomitant administration of ketoconazole, a known CYP3A4 inhibitor, with the alkaloid constituents of evodia significantly increases the area under the curve (AUC), maximum concentration (Cmax), and half-life of these constituents.

Here the concern runs in an interesting direction. Pitolisant (Wakix) acts as a mild CYP3A4 inhibitor, and the worry on file is that this kind of drug could slow how your body clears the active compounds in evodia (like evodiamine and rutaecarpine). In other words, evodia levels might build up rather than pitolisant itself being affected.

The honest truth is that this is theoretical, based only on animal and lab work using a much stronger blocker (ketoconazole), so we really don't know how it plays out in people. Evodia is also a poorly studied herb on its own. I'd skip mixing the two until you check with your pharmacist or doctor, especially since evodia's own safety isn't well established.

Theoretically, evodia might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that evodia extract inhibits hepatic CYP3A4. This effect has not been reported in humans.

Here's the situation with Wakix (pitolisant) and evodia. Your body relies on a liver enzyme called CYP3A4 to help clear pitolisant out of your system. Lab studies suggest evodia may slow that enzyme down. If that held true in people, pitolisant could build up a bit higher than expected, which might mean more of its side effects like headache, nausea, trouble sleeping, anxiety, or a faster heartbeat. Pitolisant is not a prodrug, so this points toward a stronger effect, not a weaker one.

That said, this is only based on test-tube research and has never been confirmed in humans, so I wouldn't lose sleep over it. Still, before pairing the two, check with your pharmacist or doctor first.

What Is Pitolisant Used For?

Brand name: Wakix

Wakix (pitolisant) is used to treat two hallmark problems of narcolepsy: excessive daytime sleepiness (feeling overwhelmingly tired and unable to stay awake during the day) and cataplexy (sudden episodes of muscle weakness or loss of muscle control, often triggered by emotion). It is approved for adults and children 6 years of age and older.

Both uses are specific to Wakix tablets taken by mouth. The drug is not approved for sleepiness or cataplexy caused by conditions other than narcolepsy.
