Lonafarnib falls under Cytochrome P450 2C9 (Cyp2C9) Substrates — the group Genistein Combined Polysaccharide interacts with.

Here's the short version on combining GCP with your Lonafarnib (Zokinvy). The genistein in this soy-and-mushroom supplement may nudge the activity of a liver enzyme called CYP2C9, which is one of the pathways that helps clear certain medications. If genistein slows that enzyme, a drug relying on it could build up a bit, and Lonafarnib is the kind of medicine where higher levels can mean more nausea, diarrhea, vomiting, or appetite loss.

But I want to keep this in perspective. This interaction has only been seen in animal studies so far, the rating is minor, and it's far from proven in people. Lonafarnib is mostly handled by a different enzyme (CYP3A) anyway. Still, since this drug is used for a serious condition, please loop in your prescriber and pharmacist before adding GCP.

About Lonafarnib

What Is Lonafarnib Used For?

Brand name: Zokinvy

Zokinvy (lonafarnib) is approved for patients 12 months of age and older who have a body surface area of 0.39 m² or above. It is used to reduce the risk of mortality in Hutchinson-Gilford Progeria Syndrome (HGPS) — a rare, severe premature-aging disorder.

Zokinvy is also used to treat processing-deficient Progeroid Laminopathies — a related group of rare genetic conditions — specifically those caused by a heterozygous LMNA mutation with progerin-like protein buildup, or by homozygous or compound heterozygous ZMPSTE24 mutations. It is not indicated for other Progeroid Syndromes or processing-proficient Progeroid Laminopathies.
