Olaparib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Grape interacts with.

It is unclear if grape seed extract inhibits or induces CYP3A4; research is conflicting.
In vitro evidence suggests that grape seed extract might inhibit CYP3A4 enzymes. However, evidence from animal research shows that grape seed extract may induce CYP3A4 in the liver. So far, these interactions have not been reported in humans.

Here's the short version: your body clears olaparib (Lynparza) using a liver enzyme called CYP3A4. Grape seed extract may bump into that same enzyme, but the lab and animal studies disagree on what it does. Some test-tube work suggests it slows the enzyme down (which could let olaparib build up and worsen side effects like nausea, tiredness, low blood counts, or anemia), while animal data hints it might speed the enzyme up (which could weaken olaparib's cancer-fighting effect).

The honest truth is we just don't know, and this has never actually been reported in people. Eating grapes is fine. But before adding a concentrated grape seed extract supplement, please run it by your oncologist or pharmacist first.

About Olaparib

What Is Olaparib Used For?

Brand name: Lynparza

Lynparza (olaparib) is used to treat adults with certain ovarian, fallopian tube, or primary peritoneal cancers that carry BRCA or other DNA-repair gene mutations — both as a maintenance therapy after platinum-based chemotherapy (including in combination with bevacizumab for some patients) and for recurrent disease. It is also approved for HER2-negative breast cancer with a germline BRCA mutation, both in early high-risk disease (adjuvant setting) and in metastatic disease.

Lynparza is additionally used as maintenance therapy for adults with germline BRCA-mutated metastatic pancreatic adenocarcinoma whose disease has not progressed on first-line platinum-based chemotherapy, and for adults with certain gene-mutated metastatic castration-resistant prostate cancer — either alone (after prior enzalutamide or abiraterone) or in combination with abiraterone and prednisone or prednisolone. All uses require testing for specific genetic mutations using an FDA-approved companion diagnostic before starting treatment.
