Amitriptyline, Chlordiazepoxide falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Higenamine interacts with.

Theoretically, higenamine might increase the levels and clinical effects of drugs metabolized by CYP2D6.
In vitro research shows that higenamine inhibits CYP2D6 enzymes. However, this effect has not been reported in humans.

Limbitrol DS combines amitriptyline (a tricyclic antidepressant) with chlordiazepoxide (a benzodiazepine). The amitriptyline part is cleared in good measure by an enzyme called CYP2D6. In lab studies, higenamine appears to block that enzyme, so in theory amitriptyline could build up and hit harder. That might mean more drowsiness, dry mouth, constipation, blurry vision, dizziness on standing, and in larger amounts, effects on the heart rhythm.

I'll be honest: this is only seen in test-tube research, not in real people, so it's theoretical. Still, higenamine is a heart and blood pressure stimulant with unreliable labeling, which makes it a poor mix with this particular medicine. Please run it by your pharmacist or doctor before combining them.

Theoretically, higenamine might increase the levels and clinical effects drugs metabolized by CYP3A4.
In vitro research shows that higenamine inhibits CYP3A4 enzymes by 21%. However, this effect has not been reported in humans.

Limbitrol DS combines two active medicines: amitriptyline (an older antidepressant) and chlordiazepoxide (a benzodiazepine for anxiety). Both rely partly on a liver enzyme called CYP3A4 to be cleared from your body. In lab dishes, higenamine slightly blocked that enzyme (about 21%). If that held true in people, these medicines could build up a bit and hit you harder, meaning more drowsiness, dizziness, dry mouth, constipation, or feeling foggy and unsteady.

Here's my honest take: this is only test-tube evidence and has never been confirmed in humans, so it's theoretical and likely mild. The bigger worry is higenamine itself, which acts like adrenaline and can raise heart rate and blood pressure, something amitriptyline can also affect. Please run this combination by your pharmacist or doctor before mixing them.
