Higenamine and Hydrocodone, Ibuprofen

These two interact in 3 distinct ways. Each one is explained separately below.

Why Are There 3 Interactions?

Higenamine interacts with whole categories of medication. Hydrocodone, Ibuprofen falls into 3 of those categories, so it can interact with Higenamine in 3 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

1 of 3

Why this applies to Hydrocodone, Ibuprofen

Hydrocodone, Ibuprofen falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Higenamine interacts with.

Theoretically, higenamine might increase the levels and clinical effects of drugs metabolized by CYP2D6.
In vitro research shows that higenamine inhibits CYP2D6 enzymes. However, this effect has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Here's the wrinkle with Vicoprofen, which pairs hydrocodone with ibuprofen. The hydrocodone part is actually a prodrug of sorts: your body uses an enzyme called CYP2D6 to turn some of it into hydromorphone, a stronger painkiller. Lab studies hint that higenamine may block that same enzyme. If that happened in people, you could convert less hydrocodone to its active form, which might mean weaker pain relief than expected.

Honestly, this is only seen in test-tube research, not in real patients, so I wouldn't lose sleep over it. The bigger worry is mixing a heart-stimulating supplement with any opioid. Please run this combination by your pharmacist or doctor before using both.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

2 of 3

Hydrocodone, Ibuprofen also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, higenamine might increase the levels and clinical effects drugs metabolized by CYP3A4.
In vitro research shows that higenamine inhibits CYP3A4 enzymes by 21%. However, this effect has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Vicoprofen combines hydrocodone (an opioid pain reliever) with ibuprofen. The hydrocodone part is partly handled by a liver enzyme called CYP3A4. Lab studies suggest higenamine may slow that enzyme down a little (about 21% in a test tube), which in theory could let hydrocodone build up a bit and feel stronger. That might mean more drowsiness, dizziness, nausea, constipation, or slowed breathing.

Here's the honest part: this comes only from lab data, not from real people, so we don't know if it actually matters in your body. I'd also flag that higenamine itself can raise heart rate and blood pressure, which is its own concern. Before combining them, please check with your pharmacist or doctor.

ANTICOAGULANT/ANTIPLATELET DRUGS

3 of 3

Hydrocodone, Ibuprofen also falls under ANTICOAGULANT/ANTIPLATELET DRUGS.

Theoretically, higenamine might increase the risk of bleeding or bruising when taken with anticoagulant/antiplatelet drugs.
Animal research shows that higenamine inhibits platelet aggregation and reduces the size of thrombus formation.

Let's talk about pairing higenamine with Vicoprofen, which combines hydrocodone (an opioid) with ibuprofen (an NSAID). The piece that matters here is the ibuprofen. NSAIDs like ibuprofen already nudge up your bleeding risk a little by affecting how your platelets stick together. In animal studies, higenamine also seemed to slow platelet clumping, so stacking the two could, in theory, add up to easier bruising or bleeding.

I want to be honest though: this is a theoretical concern based on lab and animal work, not strong human data. Watch for things like unusual bruising, nosebleeds, bleeding gums, or dark stools. Higenamine also stresses the heart and raises blood pressure, which is its own reason for caution. Please check with your pharmacist or doctor before combining them.

What These Ratings Mean

Every interaction above is graded three ways — here’s the key to each label.

Severity how clinically significant the interaction is
Major

Clinically significant. Avoid the combination, or use only under close professional supervision.

Moderate

Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.

Minor

Generally not serious. Be aware of it and mention it to your pharmacist.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

From our pharmacist’s overview on the Hydrocodone, Ibuprofen page

“Vicoprofen pairs hydrocodone, an opioid, with ibuprofen, an NSAID, which gives supplements two separate ways to cause trouble, and 13 of the 379 interactions on file are rated major, so this page deserves a careful read rather than a skim. The biggest patterns are bleeding and sedation: more than 200 of these ingredients can add to ibu…”

About Higenamine

What is Higenamine?

  • Weight loss and fat burning
  • Pre-workout energy and performance
  • Appetite suppression
  • Athletic stimulant
  • Traditional medicine remedies

Higenamine is a plant-based stimulant compound added to many weight-loss and pre-workout supplements, but there is very little human evidence that it works and real concerns about heart and blood pressure effects. It is banned in sports by the World Anti-Doping Agency (WADA), and product labels often list inaccurate amounts, so most people should approach it with caution and talk to a pharmacist or doctor first.
