Why this applies to Olaparib

Olaparib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Hops interacts with.

Theoretically, hops extract might alter metabolism of CYP3A4 substrates; however, this effect may not be clinically significant.
Animal research suggests that specific constituents of hops, called lupulones, can induce hepatic CYP3A4 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients with normal metabolism shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, decreases the concentration of alprazolam, a CYP3A4 probe substrate, by 7.6%. This reduction is unlikely to be clinically relevant.

Here's the short version: your body relies on a liver enzyme called CYP3A4 to break down olaparib (Lynparza). Hops may very slightly speed up that enzyme, which in theory could clear olaparib a touch faster and trim how much stays in your system. Since olaparib isn't a prodrug, faster clearance would mean slightly less drug working, not more.

But I want to keep this in perspective. This concern comes only from lab and animal studies, and a human study found a similar drug dropped by under 8 percent, which isn't enough to matter. So the real-world risk here is quite low. Still, olaparib is a serious cancer medicine, so please run any hops supplement past your oncologist or pharmacist first.

Hops are most often used for sleep and mild anxiety, frequently combined with valerian, but the human evidence is limited and not very strong. They are generally well tolerated when used short-term, though they may cause drowsiness, and should be avoided during pregnancy and breastfeeding because their safety is unknown.

About Olaparib

What Is Olaparib Used For?

Brand name: Lynparza

Lynparza (olaparib) is used to treat adults with certain ovarian, fallopian tube, or primary peritoneal cancers that carry BRCA or other DNA-repair gene mutations — both as a maintenance therapy after platinum-based chemotherapy (including in combination with bevacizumab for some patients) and for recurrent disease. It is also approved for HER2-negative breast cancer with a germline BRCA mutation, both in early high-risk disease (adjuvant setting) and in metastatic disease.

Lynparza is additionally used as maintenance therapy for adults with germline BRCA-mutated metastatic pancreatic adenocarcinoma whose disease has not progressed on first-line platinum-based chemotherapy, and for adults with certain gene-mutated metastatic castration-resistant prostate cancer — either alone (after prior enzalutamide or abiraterone) or in combination with abiraterone and prednisone or prednisolone. All uses require testing for specific genetic mutations using an FDA-approved companion diagnostic before starting treatment.
