Cyclobenzaprine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Hops interacts with.

Hops extract does not seem to affect the metabolism of CYP1A2 substrates.
In vitro research suggests that flavonoid constituents of hops inhibit CYP1A2 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, does not affect levels of caffeine, a CYP1A2 probe substrate.

Here's the good news on pairing hops with cyclobenzaprine: this is a very minor concern. Cyclobenzaprine is partly handled by a liver enzyme called CYP1A2, and lab (test tube) studies hinted that flavonoids in hops might slow that enzyme down. On paper, that could let cyclobenzaprine build up a bit and add to its usual effects, like extra drowsiness, dry mouth, or feeling foggy.

But when researchers actually tested a standardized hops extract in real people, it did not change how the body cleared a CYP1A2 probe drug. So the concern stays mostly theoretical. The bigger real-world issue is simply that both hops and cyclobenzaprine can make you sleepy, so go easy and check with your pharmacist if you notice extra grogginess.

Cyclobenzaprine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, hops extract might alter metabolism of CYP3A4 substrates; however, this effect may not be clinically significant.
Animal research suggests that specific constituents of hops, called lupulones, can induce hepatic CYP3A4 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients with normal metabolism shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, decreases the concentration of alprazolam, a CYP3A4 probe substrate, by 7.6%. This reduction is unlikely to be clinically relevant.

Here's the short version: cyclobenzaprine is partly cleared by a liver enzyme called CYP3A4, and there's a thought that hops might gently nudge that enzyme to work a little faster. If that happened, your body could clear cyclobenzaprine slightly quicker, which in theory might make it feel a touch less effective at relaxing your sore muscles. Cyclobenzaprine is not a prodrug, so it doesn't need that enzyme to switch on, it just gets broken down by it.

The honest truth is this is very weak, mostly lab and animal data. In people, a similar hops extract changed a test drug's level by under 8%, which is too small to notice. I wouldn't lose sleep over it, but since both can cause drowsiness, run it by your pharmacist.

What Is Cyclobenzaprine Used For?

Brand names: Cyclobenzaprine, Flexeril, Cycloprine

Cyclobenzaprine tablets and extended-release capsules are used alongside rest and physical therapy to relieve muscle spasm caused by acute, painful musculoskeletal conditions — easing pain, tenderness, and limited movement. This use is intended for short-term treatment only (up to 2–3 weeks).

Tonmya, the sublingual (under-the-tongue) form of cyclobenzaprine, is separately indicated for the treatment of fibromyalgia in adults. It is not the same as the oral tablet or capsule formulations.
