Acetaminophen, Aspirin, Caffeine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Hops interacts with.
In vitro research suggests that flavonoid constituents of hops inhibit CYP1A2 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, does not affect levels of caffeine, a CYP1A2 probe substrate.

Here's the good news on combining hops with your Excedrin (acetaminophen, aspirin, and caffeine): this pairing is really not a concern. The interest here is the caffeine piece. In a lab dish, certain hops flavonoids appeared to slow down CYP1A2, the liver enzyme that helps clear caffeine. On paper that could let caffeine linger a bit longer, meaning slightly more jitteriness or trouble sleeping.
But that worry didn't hold up in people. An actual human study found that a standardized hops extract did not change caffeine levels at all. So this stays a minor, unlikely interaction based mostly on test-tube data. Keep in mind hops can make you drowsy on their own, which is a separate point. CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Animal research suggests that specific constituents of hops, called lupulones, can induce hepatic CYP3A4 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients with normal metabolism shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, decreases the concentration of alprazolam, a CYP3A4 probe substrate, by 7.6%. This reduction is unlikely to be clinically relevant.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version: pairing hops with a combo product like Excedrin (acetaminophen, aspirin, and caffeine) is very unlikely to cause you trouble. The concern on file is that hops might nudge a liver enzyme called CYP3A4 to work a little faster, which could in theory clear certain drugs slightly quicker. But this is based only on lab and animal studies, plus one human study where the effect was tiny (under 8%) and judged not clinically meaningful.
On top of that, the three ingredients in this product aren't mainly handled by CYP3A4, so even that small effect probably doesn't apply much here. One real-world note: both hops and caffeine can affect alertness in opposite ways, so you may notice mixed sleepiness or jitteriness. If you take these together often, just mention it to your pharmacist to be safe.
