Abemaciclib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Horny Goat Weed interacts with.

Theoretically, horny goat weed might increase the effects and side effects of CYP3A4 substrates.
In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.

Here's the gist for Verzenio (abemaciclib). Your body relies on a liver enzyme called CYP3A4 to break this medication down. In test-tube studies, horny goat weed appears to slow that same enzyme. If that happened in people, abemaciclib could clear more slowly and build up in your system, meaning stronger effects and a higher chance of side effects like diarrhea, low blood counts, fatigue, nausea, or, less commonly, liver or clotting problems.

I want to be honest about the strength of this, though. It's only seen in lab studies, not in actual patients, so it's theoretical. But because abemaciclib is a serious cancer drug with real side effects, I'd play it safe and run any supplement past your oncologist or pharmacist before starting it.

What Is Abemaciclib Used For?

Brand name: Verzenio

Verzenio (abemaciclib) is used to treat HR-positive, HER2-negative breast cancer in adults. For early breast cancer at high risk of recurrence, Verzenio is taken alongside endocrine therapy (tamoxifen or an aromatase inhibitor) after surgery to help prevent the cancer from coming back.

For advanced or metastatic breast cancer (cancer that has spread), Verzenio can be used in combination with an aromatase inhibitor as a first hormonal treatment, in combination with fulvestrant after the cancer has progressed on endocrine therapy, or alone after prior endocrine therapy and chemotherapy have stopped working.
