Lapatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Horny Goat Weed interacts with.

Theoretically, horny goat weed might increase the effects and side effects of CYP3A4 substrates.
In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.

Here's the basic idea with lapatinib (Tykerb). Your body relies on a liver enzyme called CYP3A4 to clear this drug from your system. In lab dish studies (not in actual people), horny goat weed seemed to slow that enzyme down. If that were to happen in the body, lapatinib could build up higher than intended, which may mean more of its usual side effects like diarrhea, nausea, rash, fatigue, and in some cases effects on the liver or heart rhythm.

I want to be honest about the strength of this: it's theoretical and based only on test-tube data, never confirmed in humans. Still, lapatinib is a serious cancer medicine, so I wouldn't add this herb without first checking with your oncologist or pharmacist.

What Is Lapatinib Used For?

Brand name: Tykerb

Lapatinib tablets — sold under the brand name Tykerb — are used to treat advanced or metastatic breast cancer in patients whose tumors overexpress the HER2 protein. They are always used in combination with another medicine, not alone.

Tykerb is approved in combination with capecitabine for HER2-positive advanced or metastatic breast cancer in patients who have already received prior treatment including an anthracycline, a taxane, and trastuzumab, and whose disease has progressed on trastuzumab. Tykerb is also approved in combination with letrozole for postmenopausal women with hormone receptor-positive, HER2-positive metastatic breast cancer when hormonal therapy is appropriate.
