Here's the short version: denosumab (Prolia or XGEVA) works by quieting part of your immune signaling to slow bone breakdown, and in some uses it lowers immune activity. Lion's mane, on the other hand, has shown in lab and animal studies that its mushroom sugars (polysaccharides) may nudge the immune system to be more active. In theory, those two could pull in opposite directions and blunt some of what denosumab is doing.

I want to be honest about how thin this evidence is. This concern comes only from test-tube and animal research, not from people taking both together, so the real-world risk is likely small. Denosumab isn't a prodrug, and lion's mane doesn't change how it's cleared. Still, before adding a lion's mane supplement, run it by your doctor or pharmacist so they can weigh it for your situation.

About Denosumab Injection

What Is Denosumab Injection Used For?

Brand names: Prolia, XGEVA

Jubbonti (denosumab) injection is used to treat osteoporosis in postmenopausal women and men at high risk for fracture, including those who have already had a fracture or who have not responded to other osteoporosis medicines. It is also used to treat osteoporosis caused by long-term steroid (glucocorticoid) use in men and women.

Jubbonti is additionally used to increase bone mass in men receiving androgen deprivation therapy (a hormone-lowering treatment) for nonmetastatic prostate cancer, and in women receiving aromatase inhibitor therapy for breast cancer — both of which can weaken bones over time.
