Ixazomib Citrate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Polypodium Leucotomos interacts with.

Theoretically, Polypodium leucotomos might increase levels of drugs metabolized by CYP3A4.
In vitro and animal research suggests that Polypodium leucotomos can decrease the metabolism of midazolam by a mechanism possibly related to the inhibition of CYP3A4. However, a small study in humans suggests that taking Polypodium leucotomos with midazolam does not affect the metabolism of midazolam.

Here's the gist for Ninlaro (ixazomib): your body partly clears it using a liver enzyme called CYP3A4. The thinking is that Polypodium leucotomos might slow that enzyme a little, which could in theory let ixazomib build up and bump up its side effects, things like nausea, diarrhea or constipation, fatigue, low blood counts, or nerve tingling in the hands and feet.

That said, this is mostly a theoretical concern. The strongest human data we have, using a test drug called midazolam, showed no real change. So a meaningful interaction with ixazomib is unlikely. Still, since ixazomib is a serious cancer medicine with a tight dosing window, please run any supplement past your oncology pharmacist or doctor before starting it.

What Is Ixazomib Used For?

Brand name: Ninlaro

Ninlaro (ixazomib) is used to treat multiple myeloma — a cancer of plasma cells in the bone marrow. It is not used alone; it is always taken together with two other medicines, lenalidomide and dexamethasone. This combination is for adults who have already received at least one prior treatment for their myeloma.

Ninlaro is not recommended for use as maintenance therapy or in newly diagnosed multiple myeloma patients on a lenalidomide and dexamethasone regimen outside of controlled clinical trials.
