Animal research shows that poria mushroom extract has sedative properties. This interaction has not been shown in humans.

Here's the simple version: diphenhydramine (Benadryl) already makes a lot of people drowsy. It calms down the central nervous system, which is exactly why it's used as a sleep aid. Poria mushroom has shown mild calming, sedative-like effects in animal studies, so the concern is that stacking the two could leave you more drowsy, groggy, foggy-headed, or unsteady than you'd expect from the Benadryl alone.
I want to be honest about the evidence, though: this is theoretical. It's based on animal research and how these things work on paper, not on studies in people. So I wouldn't panic, but it's worth a heads-up. If you take both, go easy until you know how you feel, avoid driving until you're sure, and check with your pharmacist or doctor first.

In animal research, poria mushroom essential oil reduces acetylcholinesterase activity. This interaction has not been shown in humans.

Here's the gist of it. Diphenhydramine (Benadryl) works by blocking acetylcholine, the very chemical messenger that helps with things like memory, salivation, and bladder control. That blocking is what dries you up and makes you drowsy. In animal studies, poria mushroom appears to raise available acetylcholine, which could, in theory, push back against what Benadryl is doing and make it work a little less well.
So you might notice your allergy relief or sleep aid feels slightly weaker. Honestly though, this has only been seen in lab animals, never proven in people, so I wouldn't lose sleep over it. Diphenhydramine isn't a prodrug, so nothing flips here. If you regularly use both, just mention it to your pharmacist or doctor.

Most oral diphenhydramine products temporarily relieve symptoms of hay fever and other upper respiratory allergies: runny nose, sneezing, itchy and watery eyes, and an itchy nose or throat. They also temporarily relieve runny nose and sneezing from the common cold.
After Bite Xtra is a topical gel. It is used for temporary relief of pain and itching from minor burns, minor cuts or scrapes, insect bites, minor skin irritations, and rashes from poison ivy, poison oak and poison sumac.
