Why this applies to Nystatin
Nystatin falls under ANTIFUNGALS — the group Saccharomyces Boulardii interacts with.
S. boulardii is a live yeast. Therefore, simultaneously taking antifungals might kill a significant number of the organisms.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the key thing to know: this interaction runs the other way around from what people usually expect. Nystatin is an antifungal, and S. boulardii is a live, beneficial yeast. Since nystatin's whole job is to stop fungi from growing, taking the two together could kill off some of the probiotic yeast before it has a chance to work. So it's the supplement's effectiveness that may take the hit, not the nystatin's.
The good news is this concern is theoretical, based on how these two work rather than real-world reports, and it's rated as minor. You shouldn't expect new side effects from the nystatin itself. If you're using S. boulardii for something like diarrhea, separating the doses by a few hours may help. Check with your pharmacist before combining them.

Nystatin treats infections caused by Candida, a type of yeast. Which infection it treats depends on the form you use.
- Topical creams, ointment and powders (including Mycostatin, Nyamyc, Nystop and Klayesta): skin or mucous membrane Candida infections.
- Oral suspension: Candida infections in the mouth.
- Oral tablets: Candida infection of the digestive tract lining (not the esophagus).
The topical products are not for use in the mouth, vagina or eyes, and not for infections that spread through the body.
