Ethynodiol Diacetate, Mestranol falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Aegeline interacts with.

Aegeline inhibits cytochrome P450 3A4 (CYP3A4) activity in vitro. So far, this interaction has not been reported in humans. Theoretically, aegeline may increase levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others. Use aegeline cautiously or avoid in patients taking these drugs.

Ovulen is a birth control pill that combines two hormones (ethynodiol diacetate and the estrogen mestranol). Your liver uses an enzyme called CYP3A4 to break these hormones down. Aegeline has been shown to slow that same enzyme, but only in lab dish studies, not in real people. In theory, if it did slow CYP3A4 in your body, more hormone could linger and you might notice estrogen-type effects like nausea, breast tenderness, headaches, or bloating.

Honestly, the bigger concern here is aegeline itself. It has been tied to serious liver injury and regulators have warned against products containing it, so I'd steer clear of it regardless. Please check with your pharmacist or doctor before mixing any supplement with your birth control.

Aegeline is a compound originally identified in the bael tree (Aegle marmelos) that gained attention because it was added to some weight-loss and workout supplements.
