Why this applies to Lorlatinib
Lorlatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Beta-methylphenethylamine (bmpea) interacts with.
In vitro research shows that BMPEA inhibits CYP3A4 by up to 37%.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the basic idea with these two together. Your body relies on a liver enzyme called CYP3A4 to break down lorlatinib and keep its levels in a safe range. BMPEA appears to slow that enzyme down a bit (lab studies show up to about a one-third drop in activity). If that happened in real life, lorlatinib could build up higher than intended, which may worsen its known side effects like high cholesterol or triglycerides, swelling, weight gain, mood or memory changes, fatigue, and possible effects on heart rhythm or the liver.
I'll be honest: this is a theoretical concern from test-tube data, not proven in people. Still, since BMPEA is an unapproved stimulant with its own heart risks, I'd avoid it and check with your oncology team first.

Beta-methylphenethylamine (BMPEA) is a synthetic stimulant chemically similar to amphetamine that has been found in some weight-loss and sports supplements, often without being clearly listed on the label. What Is Lorlatinib Used For?
Brand name: Lorbrena
Lorbrena (lorlatinib) is used to treat adults with metastatic non-small cell lung cancer (NSCLC) — meaning lung cancer that has spread — when lab testing confirms the tumor is ALK-positive (has a specific gene change that drives cancer growth). Patients must be tested with an FDA-approved diagnostic test before starting treatment.
Lorbrena has been studied in patients who have not yet received treatment for advanced disease, as well as in patients whose cancer progressed on other ALK-targeting medicines.
