Bitter Orange + Propoxyphene
Propoxyphene brand names: Darvon, Prophene-65, Margesic, Davon.
Bitter Orange has a minor interaction with Propoxyphene. Theoretically, bitter orange might increase levels of drug metabolized by CYP2D6.

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
Why this applies to Propoxyphene
Propoxyphene falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Bitter Orange interacts with.
In vitro research shows that octopamine, a constituent of bitter orange, weakly inhibits CYP2D6 enzymes. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version on combining bitter orange with propoxyphene. Your body uses an enzyme called CYP2D6 to help clear propoxyphene. Lab studies suggest one compound in bitter orange (octopamine) might weakly block that enzyme. If that happened, propoxyphene could build up a bit, which in theory means stronger opioid effects like extra drowsiness, dizziness, constipation, or slowed breathing.
The important part: this is only seen in test-tube research, never confirmed in real people, and it's rated a minor, possible concern. So I wouldn't lose sleep over it. Still, bitter orange is a stimulant that raises heart rate and blood pressure, so it's worth a quick chat with your pharmacist or doctor before pairing the two.

From our pharmacist’s overview on the Propoxyphene page
“Propoxyphene (Darvon) is an opioid painkiller, and the biggest hazard among the 172 supplement interactions on file is anything else that slows the brain and breathing. Seventy-eight of these ingredients pile on that sedation, alcohol and kava among the worst rated, and with an opioid the combination can push breathing dangerously low.”

About Bitter Orange
What is Bitter Orange?
- Weight loss
- Athletic performance and energy
- Digestive upset
- Appetite control
- Aromatherapy for stress (neroli oil)
Bitter orange is a citrus fruit whose extracts contain synephrine, a mild stimulant often added to weight-loss and energy supplements. Evidence that it works for weight loss or performance is limited, and because it can raise heart rate and blood pressure, people with heart problems, high blood pressure, or who are pregnant should avoid it. Talk to your pharmacist or doctor before using it, especially if you take other stimulants.
About Acetaminophen, Butalbital, and Caffeine
What Is Acetaminophen, Butalbital, and Caffeine Used For?
Brand names: Darvon, Prophene-65, Margesic, Davon
This combination product — available as capsules, tablets, and oral solution under various names including Fioricet — is used to relieve the symptoms of tension headaches (also called muscle contraction headaches).
All supported products, including Fioricet capsules and the oral solution, share this same indication. Because butalbital can be habit-forming, extended or repeated use is not recommended, and these products are not established as safe or effective for treating headaches that come back frequently over time.
