Amitriptyline falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Black Hoof Mushroom interacts with.

Theoretically, black hoof mushroom might increase the levels and clinical effects of CYP1A2 substrates.
In vitro, polysaccharides from black hoof mushroom grown in culture inhibit CYP1A2. This effect has not been reported in humans.

Here's the gist: your body uses an enzyme called CYP1A2 to help clear amitriptyline. In a lab dish, compounds from black hoof mushroom slowed that enzyme down. If the same thing happened in people, amitriptyline could build up a bit and you might notice more of its usual effects, like drowsiness, dry mouth, constipation, blurry vision, or feeling lightheaded when you stand up.

I want to be honest with you though: this is only a theoretical concern based on test-tube work, and it has not been shown in actual humans. So I wouldn't lose sleep over it. Still, since amitriptyline can be sensitive to changes, it's worth a quick chat with your pharmacist or doctor before adding this mushroom.
