Tucatinib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Cannabinol (cbn) interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of cannabinol.
In vitro research shows that CYP3A4 enzymes are involved in the 8-hydroxylation of cannabinol. So far, this interaction has not been reported in humans.

Here's the key thing to know: this interaction works in one direction. Tucatinib (Tukysa) blocks an enzyme called CYP3A4, and that same enzyme helps break down the supplement CBN. So if you take CBN while on tucatinib, the CBN may clear out of your body more slowly and build up a bit. In plain terms, you might feel more of CBN's effects, like extra drowsiness or grogginess. Tucatinib itself isn't the one piling up here.

That said, this is only based on lab studies, not real people, so it's more of a theoretical caution than a proven problem. Since you're managing a serious cancer treatment, please run any CBN or cannabis product past your oncology team or pharmacist first.

What Is Tucatinib Used For?

Brand name: Tukysa

Tukysa (tucatinib) tablets are used in combination with trastuzumab and capecitabine to treat adults with advanced, unresectable, or metastatic HER2-positive breast cancer — including those whose cancer has spread to the brain — who have already received at least one HER2-targeted treatment.

Tukysa tablets are also used in combination with trastuzumab alone to treat adults with RAS wild-type, HER2-positive unresectable or metastatic colorectal cancer that has continued to grow after prior chemotherapy. This colorectal cancer use has received accelerated approval, meaning final approval depends on further study results confirming the benefit.
