Evodia might reduce the levels and clinical effects of CYP1A2 substrates through induction of CYP1A2.
Evodia extract and the evodia constituent rutaecarpine induce hepatic CYP1A2 enzyme activity. Evodia decreases levels of theophylline and caffeine, CYP1A2 substrates, by about 70% in animal models.

Here's the gist for Zileuton (Zyflo). Your liver uses an enzyme called CYP1A2 to clear this drug. Evodia appears to speed that enzyme up, so in theory it could push Zileuton out of your system faster and leave less of it working. For an asthma controller medicine, weaker levels could mean it does a poorer job preventing wheezing, coughing, chest tightness, and shortness of breath.

I want to keep this in perspective. This concern comes from lab and animal research, not solid human studies, so we can't say how big the effect would be in real people. Still, since Zileuton keeps your airways calm day to day, I wouldn't combine it with concentrated Evodia supplements without checking with your pharmacist or doctor first.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of evodia.
Animal research shows that concomitant administration of ketoconazole, a known CYP3A4 inhibitor, with the alkaloid constituents of evodia significantly increases the area under the curve (AUC), maximum concentration (Cmax), and half-life of these constituents.

Here the concern runs the opposite way from what most people expect. Zileuton isn't the drug that builds up. Instead, your asthma medicine can act as a mild blocker of a liver enzyme called CYP3A4, which is one of the pathways that helps clear Evodia's active compounds (evodiamine and rutaecarpine) from your body. So in theory, taking them together could let Evodia hang around longer and reach higher levels, possibly making its effects and any side effects stronger.

The good news: this is only suggested by lab and animal studies, not real human cases, so it's far from certain. Even so, since Evodia itself is poorly studied, I'd run it past your pharmacist or doctor before combining the two, especially while your asthma is being managed.

Here's the thing to understand about this particular pairing: the concern runs toward the Evodia, not your Zileuton (Zyflo). Zileuton happens to slow down a liver enzyme called CYP1A2, and that same enzyme helps break down one of Evodia's active compounds (rutaecarpine). So in theory, taking them together could let the Evodia build up a bit higher than expected, which might make any of its effects or side effects stronger.

I want to keep this in perspective, though. This is based only on lab and animal research, so it's a theoretical worry, not something proven in people. Your asthma control from Zileuton shouldn't change. Still, since Evodia isn't well studied and your Zileuton already needs liver monitoring, please run this combination past your pharmacist or doctor first.

Here's the basic idea with Evodia and Zileuton. Your body uses a liver enzyme called CYP3A4 to help clear zileuton out. In test-tube studies, evodia seems to slow that enzyme down. If that happened in real life, zileuton could build up a bit higher than usual, which might mean more of its typical side effects, like nausea, stomach upset, headache, or, less commonly, signs of liver irritation.

I want to be honest about how shaky this is. This warning comes only from lab research, not from people, so we really don't know if it matters in a real body. Zileuton is not a prodrug, so the direction (slightly stronger effect) makes sense here. Still, before pairing them, run it by your pharmacist or doctor first.

About Zileuton

What Is Zileuton Used For?

Brand names: Zyflo, Zyflo CR

Zileuton is used for the ongoing prevention and treatment of asthma in adults and adolescents aged 12 and older. It is a maintenance medicine — meaning it is taken regularly to keep asthma under control, not to stop an asthma attack once it has started.

Zyflo (the tablet form) and zileuton extended-release tablets are both approved for this same purpose. Neither form should be used to treat a sudden asthma attack. However, both can be continued during a flare-up of asthma while you also use your rescue inhaler.
