Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Fennel interacts with.

Theoretically, fennel might increase levels of drugs metabolized by CYP3A4.

In vitro research suggests that fennel inhibits CYP3A4 enzyme activity. This effect has not been reported in humans.

Your body relies on a liver enzyme called CYP3A4 to clear entrectinib (Rozlytrek). In a test tube, fennel appeared to slow that enzyme down. If the same thing happened in people, entrectinib could build up a bit higher than expected, which might mean more of its usual side effects, things like dizziness, fatigue, weight gain, swelling, constipation, or changes in heart rhythm.

I want to be honest about the strength of this, though. This comes only from lab studies and has not been seen in actual patients. Everyday culinary fennel in cooking or tea is very unlikely to be a problem. Concentrated fennel capsules or fennel essential oil are where I'd pause. Since entrectinib is a serious cancer medicine, please run any fennel supplement past your oncology team or pharmacist first.

Fennel is a Mediterranean herb widely used as a food and spice, and traditionally taken for digestive complaints, colic, and menstrual cramps. Some small studies suggest possible benefit for certain stomach and menstrual symptoms, but the evidence is limited. What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC) — a type of lung cancer where the ROS1 gene is rearranged — as confirmed by an FDA-approved test.

Rozlytrek is also approved for adult and pediatric patients older than 1 month with solid tumors that have an NTRK gene fusion (a specific genetic alteration), detected by an FDA-approved test, that have spread or cannot be removed without causing serious harm, and have worsened after prior treatment or have no good treatment alternatives. This NTRK indication was granted accelerated approval based on tumor response rates; continued approval may depend on results from confirmatory trials.
