Acetaminophen, Methocarbamol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Grape interacts with.
A small pharmacokinetic study in healthy adults shows that ingestion of 200 mL of grape juice decreases phenacetin plasma levels. This is thought to be due to induction of CYP1A2.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Robaxacet (acetaminophen plus methocarbamol) and grape. One small study suggested grape juice might rev up a liver enzyme called CYP1A2, which could in theory clear certain medicines a little faster. If that nudged your levels down, the most realistic effect would be slightly less pain or muscle-relaxing benefit, not a safety scare. This is theoretical and based on weak evidence, so I wouldn't lose sleep over it.
Honestly, the bigger thing with acetaminophen is total daily dose and your liver, so be mindful of how much you take and any alcohol. A regular serving of grapes or juice is very unlikely to matter. If you're using concentrated grape seed extract, just run it by your pharmacist or doctor first.

CYTOCHROME P450 2E1 (CYP2E1) SUBSTRATES

In vitro and animal research suggests that grape seed proanthocyanidin extract inhibits CYP2E1 enzymes. However, this interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Robaxacet (which combines acetaminophen and methocarbamol) and grape. The acetaminophen part is partly handled by a liver enzyme called CYP2E1. Lab and animal studies hint that concentrated grape seed extract might slow that enzyme down. If that happened in people, acetaminophen could clear a little slower and hang around longer.
The reassuring part: this has only been seen in test tubes and animals, never reported in real patients, so I wouldn't lose sleep over it. Eating grapes or drinking grape juice almost certainly won't matter. Where I'd be a bit more thoughtful is with high-dose grape seed supplements taken alongside acetaminophen, since acetaminophen is hard on the liver in large amounts. Keep your acetaminophen within recommended limits, and check with your pharmacist before starting a grape supplement.
