Why this applies to Naproxen, Sumatriptan

Naproxen, Sumatriptan falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Grape interacts with.

Theoretically, grape juice might reduce the levels of CYP1A2 substrates.
A small pharmacokinetic study in healthy adults shows that ingestion of 200 mL of grape juice decreases phenacetin plasma levels. This is thought to be due to induction of CYP1A2.

Treximet combines naproxen (an anti-inflammatory pain reliever) with sumatriptan (a migraine medicine). The concern here is that grape juice may gently speed up an enzyme called CYP1A2, which helps your body process certain drugs. If that enzyme works faster, a medication that depends on it could clear out a bit quicker and feel less effective.

Honestly, this one is mostly theoretical and based on a small study using a different drug, so I wouldn't lose sleep over it. Neither naproxen nor sumatriptan relies heavily on CYP1A2, so a real-world drop in your migraine relief is unlikely. Eating grapes or drinking juice with your normal meals should be fine. If you take a concentrated grape supplement regularly, just mention it to your pharmacist or doctor.

Treximet combines naproxen (an NSAID) with sumatriptan (a migraine medicine). The piece that matters here is the naproxen. Like other NSAIDs, naproxen already makes your platelets a little less sticky, so it can raise your chances of bleeding or bruising, and it can be hard on the stomach. Grape supplements may have a mild blood-thinning effect of their own, so stacking the two could, in theory, add up.

Here's the honest part: this concern comes only from lab studies, not real patients, so it's pretty theoretical. Eating grapes or raisins won't be a problem. With concentrated grape seed extract, it's worth a quick chat with your pharmacist, especially if you notice easy bruising, nosebleeds, or dark stools.

It is unclear if grape juice or grape seed extract inhibits CYP2C9; research is conflicting.
In vitro evidence shows that grape seed extract or grape juice might inhibit CYP2C9 enzymes. However, a small pharmacokinetic study in healthy adults shows that drinking 8 ounces of grape juice once does not affect the clearance of flurbiprofen, a probe-drug for CYP2C9 metabolism. The effects of continued grape juice consumption are unclear.

Treximet combines naproxen (an anti-inflammatory pain reliever) with sumatriptan (a migraine medication). The naproxen part is partly cleared by a liver enzyme called CYP2C9. Some lab and test-tube studies suggest grape seed extract or grape juice might slow this enzyme down a little. If that happened, naproxen could linger longer in your body, which in theory might mean more stomach upset, heartburn, or a higher chance of stomach irritation.

Here's the reassuring part: this is weak, conflicting evidence. In one human study, an 8-ounce glass of grape juice didn't change how a similar drug was cleared. Enjoying grapes as food is unlikely to matter. If you're considering a concentrated grape seed supplement, just run it by your pharmacist or doctor first.
