Imipramine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Higenamine interacts with.

Theoretically, higenamine might increase the levels and clinical effects of drugs metabolized by CYP2D6.
In vitro research shows that higenamine inhibits CYP2D6 enzymes. However, this effect has not been reported in humans.

Here's the short version for imipramine (Tofranil). Your body relies on a liver enzyme called CYP2D6 to clear this tricyclic antidepressant out of your system. In test-tube studies, higenamine appears to slow that same enzyme down. If that happened in real life, imipramine could build up to higher levels than expected, which can mean more side effects like a fast or irregular heartbeat, dizziness, drowsiness, dry mouth, constipation, or trouble urinating.

I want to be honest about the evidence: this is theoretical and only seen in lab studies, not in actual people. On top of that, higenamine itself can rev up heart rate and blood pressure, which doesn't pair well with imipramine. Please check with your pharmacist or doctor before combining them.

Theoretically, higenamine might increase the levels and clinical effects drugs metabolized by CYP3A4.
In vitro research shows that higenamine inhibits CYP3A4 enzymes by 21%. However, this effect has not been reported in humans.

Here's the gist: your body uses a liver enzyme called CYP3A4 to help clear imipramine (Tofranil). Higenamine has been shown, in test-tube studies only, to slow that enzyme down a bit (about 21%). In theory, that could let more imipramine build up in your system, which might mean stronger effects and more side effects like a dry mouth, constipation, drowsiness, dizziness on standing, and a faster or irregular heartbeat.

I want to be honest though: this has not actually been seen in people, so it's a theoretical concern. What worries me more is that higenamine itself is a heart and blood pressure stimulant, and imipramine can already affect heart rhythm. Stacking the two could be hard on your heart. Please run this by your pharmacist or doctor before combining them.

Higenamine is a plant-based stimulant compound added to many weight-loss and pre-workout supplements, but there is very little human evidence that it works and real concerns about heart and blood pressure effects. What Is Imipramine Used For?

Brand names: Tofranil, Tofranil PM

Imipramine is used to relieve symptoms of depression. Both Imipramine Hydrochloride tablets and Imipramine Pamoate capsules are approved for this use. Endogenous depression — the kind that seems to arise from within rather than a clear outside cause — tends to respond especially well. It may take one to three weeks before you notice the full benefit.

Imipramine Hydrochloride tablets are also approved as a short-term, add-on treatment to help reduce nighttime bedwetting (nocturnal enuresis) in children aged 6 and older, after a doctor has ruled out physical causes.
