Why this applies to Vemurafenib

Vemurafenib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Horny Goat Weed interacts with.

Theoretically, horny goat weed might increase the effects and side effects of CYP3A4 substrates.
In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.

Here's the basic idea with vemurafenib (Zelboraf). Your body uses a liver enzyme called CYP3A4 to help clear this cancer medicine. In test-tube studies, horny goat weed appears to slow that same enzyme down. If that were to happen in people, your body might clear vemurafenib more slowly, letting levels creep up and possibly making side effects worse, things like joint pain, rash and sun-sensitive skin, fatigue, or changes in heart rhythm (QT prolongation).

I want to be honest, though: this is only a theoretical concern from lab work, and it has not been shown in actual patients. Still, vemurafenib is serious medicine for a serious illness, so please don't add horny goat weed without first checking with your oncologist or pharmacist.

About Vemurafenib

What Is Vemurafenib Used For?

Brand name: Zelboraf

Zelboraf (vemurafenib) is used to treat adults with unresectable (cannot be surgically removed) or metastatic (spread to other parts of the body) melanoma whose tumors have a specific genetic change called the BRAF V600E mutation, confirmed by an FDA-approved test. It is not for melanoma without a BRAF mutation (wild-type BRAF melanoma).

Zelboraf is also used to treat Erdheim-Chester Disease (ECD), a rare condition where abnormal immune cells build up in organs and tissues, in patients whose disease carries a BRAF V600 mutation.
