Here's the gist: amitriptyline already carries a sedating punch. Drowsiness, grogginess, and a foggy or heavy feeling are some of its most common side effects, especially when you first start it or take it at higher doses. Poria mushroom has shown mild calming, sedative-like activity in animal studies, so the theory is that pairing the two could stack on extra sleepiness.
I want to be honest with you, though: this concern is theoretical and based on lab and animal research, not anything proven in people. It's not a reason to panic. Just be a little extra cautious about feeling more tired than usual, and don't drive until you know how the combination affects you. Since amitriptyline is a strong sedative on its own, run this past your pharmacist or doctor before adding poria.

In animal research, poria mushroom essential oil reduces acetylcholinesterase activity. This interaction has not been shown in humans.

Here's the simple version. Amitriptyline (Elavil) has what we call anticholinergic activity, which is partly why it can cause dry mouth, blurry vision, constipation, and grogginess. Lab work in animals hints that poria mushroom may nudge things in the opposite direction by raising acetylcholine levels. In theory, that could slightly blunt amitriptyline's anticholinergic side effects, and possibly some of its intended action too.
The honest truth is that this is theoretical only. It has not been shown in people, so I wouldn't lose sleep over it. Still, since amitriptyline is a sensitive medication, it's worth a quick chat with your pharmacist or doctor before you add poria, so they can keep an eye on how you're feeling.
