In animal research, intraperitoneal rauwolscine lowered the seizure threshold level of the drug metrazol.

Here's the gist for your cough product (the one that pairs guaifenesin with hydrocodone, an opioid). Both opioids like hydrocodone and the stimulant rauwolscine can, in certain situations, nudge the brain toward being more prone to seizures. Stacking the two together might, in theory, add up and slightly raise that risk. I want to be honest though: this concern comes from old animal research where rauwolscine lowered the seizure threshold for a lab chemical, not from human studies, so the evidence is weak.
For most people this won't be a problem, but rauwolscine can also crank up heart rate, blood pressure, and anxiety, which isn't a great mix with a sedating opioid. If you have a seizure history or heart issues, please run this combo by your pharmacist or doctor first.

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

Rauwolscine is structurally related to yohimbine. In vitro research shows that yohimbine inhibits CYP2D6 enzyme activity.

Here's the wrinkle with this combo: hydrocodone is actually a prodrug in part. Your body uses an enzyme called CYP2D6 to convert some of it into hydromorphone, a more powerful painkiller. Rauwolscine (a cousin of yohimbine) may block that enzyme. So instead of building up, the strong active form could form less efficiently, which in theory means you might get a bit less pain and cough relief from a given dose. The guaifenesin part isn't affected here.
Honestly, this is only a theoretical concern based on lab data, not real patient reports, so I wouldn't panic. The bigger practical issue is that rauwolscine is a stimulant that raises heart rate and blood pressure, while hydrocodone is a sedating opioid, so they pull against each other. Please run this past your pharmacist or doctor before mixing them.
