Berotralstat Hydrochloride falls under P-GLYCOPROTEIN SUBSTRATES — the group Sesame interacts with.

Theoretically, sesame might alter the transport of P-glycoprotein substrates.
In vitro research suggests that sesamin, a constituent of sesame, can inhibit the multi-drug transporter protein, P-glycoprotein. However, this interaction has not been reported in humans.

Here's the gist for Orladeyo (berotralstat), which is taken daily to prevent hereditary angioedema attacks. Berotralstat is carried around the body partly by a transport protein called P-glycoprotein. A compound in sesame called sesamin has been shown, in test-tube studies only, to slow down that transporter. If that effect happened in real life, slightly more berotralstat could stay in your system, which in theory might nudge up its usual side effects like stomach pain, diarrhea, heartburn, or headache.

The honest takeaway: this is theoretical, based on lab work, not on people. Sesame as a food, like seeds or oil on your meal, is very unlikely to cause any problem. If you're considering concentrated sesame supplements, just run it by your pharmacist or doctor first.
