Why this applies to Abemaciclib

Abemaciclib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sweet Annie interacts with.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Here's the situation with Sweet Annie and your Verzenio (abemaciclib). Your body relies on a liver enzyme called CYP3A4 to break down abemaciclib. Sweet Annie may tug on that enzyme, but honestly the lab data points in both directions. One compound in it (artemisinin) seems to speed CYP3A4 up, while the whole-plant extract seems to slow it down. So we can't say cleanly which way it would go in a real person.

Why it matters: if the enzyme slows, abemaciclib can build up and worsen side effects like diarrhea, fatigue, low blood counts, or nausea. If it speeds up, the drug may not work as well. This is only test-tube evidence, so don't panic, but please run it by your oncologist or pharmacist before combining them.

What Is Abemaciclib Used For?

Brand name: Verzenio

Verzenio (abemaciclib) is used to treat HR-positive, HER2-negative breast cancer in adults. For early breast cancer at high risk of recurrence, Verzenio is taken alongside endocrine therapy (tamoxifen or an aromatase inhibitor) after surgery to help prevent the cancer from coming back.

For advanced or metastatic breast cancer (cancer that has spread), Verzenio can be used in combination with an aromatase inhibitor as a first hormonal treatment, in combination with fulvestrant after the cancer has progressed on endocrine therapy, or alone after prior endocrine therapy and chemotherapy have stopped working.
