Here's the gist for this combo. Your pill, Enovid-E, pairs an estrogen (mestranol) with a progestin (norethynodrel), and like many birth control hormones, it's processed by your liver. Sweet Annie is an herb that some reports suggest may be hard on the liver too. So the worry here isn't about your hormones getting stronger or weaker, it's that two things that lean on the liver might add up and stress it more than either alone.

I want to be honest about how shaky this evidence is though. This is theoretical and based mostly on anecdotes, not solid studies, so I wouldn't lose sleep over it. Still, watch for signs like yellowing skin or eyes, dark urine, belly pain, or unusual tiredness, and check with your pharmacist or doctor before combining them.

Mestranol, Norethynodrel also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Enovid-E is an older birth control pill that combines an estrogen (mestranol) with a progestin (norethynodrel). Both hormones are processed by a liver enzyme called CYP3A4, so anything that changes that enzyme could change your hormone levels. Sweet Annie is a bit of a wild card here. In lab studies its main compound, artemisinin, seemed to speed up CYP3A4, while the whole-plant extract seemed to slow it down. So the real-world direction is genuinely unclear.

If it sped up the enzyme, your hormone levels could drop, which can mean breakthrough bleeding or, in theory, less reliable protection. This is only test-tube evidence, not human studies, so I wouldn't panic. Still, before combining them, please check with your pharmacist or doctor.
