Siponimod falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sweet Annie interacts with.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Here's the situation with Sweet Annie and Mayzent (siponimod). Your body uses a liver enzyme called CYP3A4 to help clear siponimod. Sweet Annie's main compound, artemisinin, has been shown in lab dish studies to possibly speed up that enzyme, while the whole-plant extract seemed to slow it down. In plain terms, the science is mixed and only comes from test-tube work, not from people, so we honestly can't say which direction it would go in your body.

If clearance sped up, siponimod could work less well, meaning less protection against MS relapses. If it slowed down, you might see more side effects like a slow heart rate, headache, or higher blood pressure. Because this is unproven and siponimod matters too much to gamble with, please check with your pharmacist or doctor before combining them.

Mayzent (siponimod) is approved for adults with relapsing forms of multiple sclerosis (MS). This includes three specific types: clinically isolated syndrome (a first episode of MS-like symptoms), relapsing-remitting MS (periods of attacks followed by recovery), and active secondary progressive MS (a phase where the disease gradually worsens but still has active relapses).

It is taken by mouth once daily and is intended as a long-term treatment to reduce relapses and slow disease progression.
