Here is the interaction we found between these two, with the evidence behind it.
CNS DEPRESSANTS
Why this applies to Diphenhydramine
Diphenhydramine falls under CNS DEPRESSANTS — the group Theacrine interacts with.
Animal research shows that low doses of theacrine have sedating effects, whereas high doses might have stimulant effects. Depending on the dose of theacrine used, it might increase or decrease the effects of CNS depressants. However, these effects have not yet been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the honest picture on mixing theacrine with diphenhydramine (Benadryl). Diphenhydramine is a sedating antihistamine, so it tends to make you drowsy, foggy, and a little slow. Theacrine is a caffeine-cousin used for energy and focus. On paper, the two pull in opposite directions, and the worry is they could push each other around.
The catch is that this is purely theoretical, coming from animal studies, not real human reports. In animals, low doses of theacrine seemed calming while higher doses acted like a stimulant. So depending on the dose, it might add to Benadryl's drowsiness or partly fight it. Nothing dramatic has been shown in people. Still, if you're using either one to sleep or for allergies, check with your pharmacist before combining them.

From our pharmacist’s overview on the Diphenhydramine page
“Diphenhydramine (Benadryl, Simply Sleep) is sedating by design, and most of the 147 supplements and herbal ingredients on this list lean into that. Seventy-eight are sedating substances themselves, alcohol and kava the standouts, and stacking them can slow breathing and reflexes well beyond ordinary drowsiness.”

- Caffeine-like stimulation with possibly less tolerance
Theacrine is a caffeine-like compound found naturally in certain tea plants and is sold in supplements for energy, focus, and mood.
