Avapritinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Thunder God Vine interacts with.

Theoretically, thunder god vine might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that triptolide, a constituent of thunder god vine, inhibits CYP3A4 enzymes in human microsomes. This interaction has not been reported in humans.

Here's the gist for anyone taking Avapritinib (Ayvakit) alongside Thunder God Vine. Your body clears avapritinib mainly through a liver enzyme called CYP3A4. Lab studies suggest one of the herb's compounds, triptolide, may slow that enzyme down. If that happens in real life, avapritinib could build up to higher levels than intended.

Avapritinib isn't a prodrug, so more drug in your system generally means stronger effects and more side effects, things like swelling, fatigue, low blood counts, dizziness, or memory and mood changes. I want to be honest though: this concern comes only from test-tube research and hasn't been shown in actual patients, so it's theoretical. Still, since avapritinib is a serious cancer medicine, please loop in your oncologist or pharmacist before combining the two.

About Avapritinib

What Is Avapritinib Used For?

Brand name: Ayvakit

Ayvakit (avapritinib) is approved to treat adults with unresectable (cannot be surgically removed) or metastatic (spread to other parts of the body) gastrointestinal stromal tumors (GIST) that have a specific genetic mutation called a PDGFRA exon 18 mutation, including the D842V variant. Patients must be tested for this mutation before starting treatment.

Ayvakit is also approved to treat advanced systemic mastocytosis (AdvSM) — a serious condition where abnormal mast cells build up in organs — including aggressive systemic mastocytosis, systemic mastocytosis with an associated blood cancer, and mast cell leukemia. Additionally, Ayvakit treats indolent systemic mastocytosis (ISM), a less severe form of the disease. It is not recommended for either form of mastocytosis if platelet counts are too low (below 50 × 10⁹/L).
