Estro One contains 4 active ingredients. The first three — 3,3'-diindolylmethane, indole-3-carbinol, and agaricus bisporus mushroom extract — are compounds extracted from cruciferous vegetables and mushrooms, historically used in women's health formulas.
The fourth ingredient, 5,7-dihydroxy-4'-methoxyflavone, is listed but we hold no detailed information on it. The product also contains inactive ingredients (maltodextrin, gelatin, titanium dioxide, FD&C Blue #1, magnesium stearate, and silicon dioxide) as capsule binders and fillers.

The evidence for what this product can do isn't yet established in the data we hold. Both diindolylmethane and indole-3-carbinol have been studied for breast cancer, colorectal cancer, cervical dysplasia, and related conditions — but so far the research is insufficient to rate whether they're effective for any of these.
The agaricus mushroom extract shows the same pattern across allergic rhinitis, osteoporosis, cancer, heart disease, and chemotherapy side effects. That doesn't mean the product doesn't work; it means the clinical evidence isn't conclusive enough yet to say.
How safe is it?
Well-documented data
Both diindolylmethane and indole-3-carbinol are generally well tolerated in short-term studies, but long-term safety isn't well established. The most common side effects are gastrointestinal — diarrhea, gas, nausea, and headache.
At higher doses, indole-3-carbinol has been linked to unsteadiness and tremor in a small number of people; these resolved when doses were lowered. Agaricus mushroom is generally well tolerated as a food, but supplement safety isn't well documented — rare cases of severe liver damage have been reported, mostly in people with cancer receiving chemotherapy.
For pregnancy, diindolylmethane is rated likely safe. Indole-3-carbinol has no pregnancy data on file — there isn't enough information to know either way.
Agaricus mushroom should be avoided in pregnancy due to insufficient safety data. For breastfeeding, the safety data advises against both diindolylmethane and agaricus mushroom, and indole-3-carbinol data is insufficient — talk with your doctor or pharmacist before using this product if you're nursing.

Indole-3-Carbinol
Anticoagulant/Antiplatelet Drugs
Theoretically, indole-3-carbinol might increase the risk of bleeding when used with antiplatelet or anticoagulant drugs.
In vitro research shows that indole-3-carbinol inhibits platelet aggregation.
Cytochrome P450 1A2 (Cyp1A2) Substrates
Theoretically, indole-3-carbinol might increase the metabolism of CYP1A2 substrates and lower serum concentrations.
Animal research shows that indole-3-carbinol induces CYP1A2 enzymes.
Estrogens
Indole-3-carbinol might interfere with the effects of estrogen therapy.
Preliminary clinical and in vitro evidence shows that indole-3-carbinol has antiestrogenic activity.
3,3'-Diindolylmethane
Diuretic Drugs
Theoretically, diindolylmethane might increase the risk of hyponatremia if used with sodium-depleting diuretics.
Large doses of diindolylmethane (600 mg daily) have been associated with two cases of asymptomatic hyponatremia in clinical research.
Estrogens
Theoretically, diindolylmethane might increase or decrease the effects of estrogens.
Diindolylmethane might have mild estrogenic or antiestrogenic effects. Theoretically, large amounts of diindolylmethane might interfere with hormone replacement therapy.
Cytochrome P450 1A2 (Cyp1A2) Substrates
Theoretically, diindolylmethane might lower serum levels of CYP1A2 substrates.
In vitro evidence suggests that diindolylmethane can induce CYP1A2. Theoretically, it might increase metabolism of CYP1A2 substrates and lower serum concentrations. This interaction has not been reported in humans.
Agaricus bisporus extract
Antidiabetes Drugs
Theoretically, taking agaricus mushroom with antidiabetes drugs might increase the risk of hypoglycemia.
In one clinical study in patients with type 2 diabetes who are stabilized on conventional oral hypoglycemic agents, 3 of 29 patients taking an agaricus mushroom extract 500 mg three times daily for 12 weeks reported hypoglycemia, compared to one of 29 patients in the placebo group.
