Amitriptyline, Perphenazine falls under CNS DEPRESSANTS — the group Adrue interacts with.

Animal research suggests that taking adrue in combination with sodium thiopental or diazepam increases total sleep time up to four-fold compared to the effects of the drugs alone. Theoretically, concomitant use of adrue with CNS depressants might cause additive sedation. Some CNS depressants include benzodiazepines, such as diazepam (Valium), alprazolam (Xanax), triazolam (Halcion), and estazolam (ProSom); barbiturates, such as mephobarbital (Mebaral), phenobarbital (Luminal Sodium), and pentobarbital sodium (Nembutal); zolpidem (Ambien); and others.

Here's the simple version: the combo pill you're asking about pairs amitriptyline (an older antidepressant) with perphenazine (an antipsychotic). Both of these already tend to make people drowsy on their own. Adrue is a traditional plant remedy that's also thought to have a calming, sedative quality. When you layer two sedating things on top of each other, the tiredness can add up.

So the realistic worry here is extra drowsiness: feeling foggy, dizzy, slow to react, or extra sleepy. That matters most if you drive or operate machinery. Honestly, the evidence is weak, coming only from animal studies where sleep time went up, so this is more theoretical than proven. Still, it's smart to check with your pharmacist or doctor before combining them.
