Sotorasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Beta-methylphenethylamine (bmpea) interacts with.

Theoretically, BMPEA might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that BMPEA inhibits CYP3A4 by up to 37%.

Here's the gist with sotorasib (Lumakras) and BMPEA. Your liver uses an enzyme called CYP3A4 to help break sotorasib down. In lab tests, BMPEA slowed that enzyme by roughly a third. If that happened in the body, sotorasib could clear more slowly and build up, which might mean stronger side effects like nausea, diarrhea, fatigue, or changes in your liver blood tests.

I'll be honest though, this is a theoretical concern based on test-tube data, not real patients, so we don't know if it truly matters in people. That said, BMPEA is a risky, often-hidden stimulant I'd steer clear of anyway, especially during cancer treatment. Please loop in your oncologist or pharmacist before mixing the two.

Beta-methylphenethylamine (BMPEA) is a synthetic stimulant chemically similar to amphetamine that has been found in some weight-loss and sports supplements, often without being clearly listed on the label. Because of safety concerns and the lack of human research, it is best avoided.

What Is Sotorasib Used For?

Brand name: Lumakras

Lumakras is used to treat adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have already received at least one prior cancer treatment. This use was approved under the FDA's accelerated approval pathway based on tumor response rates, and continued approval may depend on results from confirmatory trials.

Lumakras is also used in combination with panitumumab to treat adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) who have previously received certain chemotherapy regimens. In both cases, patients must first be confirmed to carry the KRAS G12C mutation using an FDA-approved test.
