Cannabinol (cbn) and Metformin, Rosiglitazone

Here is the interaction we found between these two, with the evidence behind it.

Moderate

CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES

Why this applies to Metformin, Rosiglitazone

Metformin, Rosiglitazone falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Cannabinol (cbn) interacts with.

Theoretically, cannabinol might increase the levels and clinical effects of CYP2C9 substrates.
In vitro research shows that cannabinol potently inhibits CYP2C9 enzymes. So far, this interaction has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger explains PharmD, Clinical Pharmacist

If you take Avandamet (which combines metformin and rosiglitazone), here's the piece that matters: the rosiglitazone part is partly cleared by a liver enzyme called CYP2C9. In lab dish studies, CBN appears to block that enzyme. In plain terms, that could slow how your body clears rosiglitazone, letting it build up a bit. The most realistic concern would be your blood sugar dropping too low (shakiness, sweating, dizziness) or more fluid retention and swelling, which rosiglitazone can already cause. Metformin itself isn't really affected this way.

Keep this in perspective: this is based only on test-tube research and hasn't been seen in real people. Still, since blood sugar control is involved, it's worth running CBN by your pharmacist or doctor first.
