Why this applies to Palovarotene
Palovarotene falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Cannabis interacts with.
In vitro research shows that cannabis can inhibit the activity of CYP3A4 enzymes, which might decrease the metabolism of CYP3A4 substrates. In vitro research also shows that cannabis extracts modestly inhibit the CYP3A4 metabolism of testosterone; extracts providing the specific cannabinoids CBD and cannabigerol (CBG) had stronger inhibitory effects than extracts containing THC and CBD.

Here's the gist for Palovarotene (Sohonos), the retinoid used for fibrodysplasia ossificans progressiva (FOP). Your liver clears this medicine using an enzyme called CYP3A4. Lab studies suggest that cannabis, especially CBD-heavy products, can partly block that same enzyme. In theory, that could let Palovarotene build up higher than expected in your body.
Because Palovarotene is a retinoid (think of it as vitamin A on steroids), more of it in your system could mean more of its typical downsides: very dry or peeling skin and lips, dry eyes, headaches, joint or muscle aches, and higher blood fats. I want to stress this is theoretical, based only on test-tube work, not real patients. Still, since this drug is carefully dosed, please check with your doctor or pharmacist before combining them.

What Is Palovarotene Used For?
Brand name: Sohonos
Sohonos (palovarotene) is used to reduce the volume of new heterotopic ossification — abnormal bone that forms in muscles and soft tissue — in people with fibrodysplasia ossificans progressiva (FOP). It is approved for adults and for children aged 8 years and older (females) and 10 years and older (males).
FOP is a rare genetic condition where the body progressively forms extra bone outside the skeleton, especially after injury or flare-ups. Sohonos is taken both as a daily maintenance dose and at higher doses when a flare-up occurs.
