Oestrogen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Neem interacts with.

Theoretically, neem leaf extract might increase the levels and clinical effects of CYP1A2 substrates.
In vitro research shows that neem leaf extract inhibits CYP1A2 enzymes. So far, this reaction has not been reported in humans.

Here's the gist with neem and your estrogen: your body uses a liver enzyme called CYP1A2 to help break estrogen down. In test-tube studies, neem leaf extract appears to slow that enzyme. If it slowed it in people too, estrogen could clear a little more slowly and build up, which might mean stronger effects or more of the usual estrogen side effects like breast tenderness, bloating, nausea, headaches, or mood changes.

The important part: this is only seen in lab dishes, not in real people, so it's a theoretical concern. I wouldn't lose sleep over it, but since neem itself can be risky to swallow, please run it by your pharmacist or doctor before starting it alongside your estrogen.

Theoretically, neem leaf extract might increase the levels and clinical effects of P-glycoprotein substrates.
In vitro research shows that neem leaf methanol extract inhibits renal P-glycoprotein transport activity. So far, this reaction has not been reported in humans.

Here's the gist for estrogen and neem. Your body uses a little cellular pump called P-glycoprotein to move many medicines, including estrogen, in and out of cells. In lab dish studies, neem leaf extract seemed to slow that pump down. If that happened in a real person, slightly more estrogen could hang around, which in theory might mean more of estrogen's usual effects, like breast tenderness, bloating, nausea, headaches, or spotting.

The important part: this is only seen in test tubes and animals so far, never reported in actual people, so I wouldn't lose sleep over it. Still, before pairing neem supplements with your estrogen, it's worth a quick chat with your pharmacist or doctor. Don't change anything on your own.

Here's the short version: your body uses an enzyme called CYP3A4 to break down estrogen. In lab dish studies, neem leaf extract seems to slow that enzyme down. If that happened in real life, less estrogen would get cleared, so more could build up in your system. That might mean stronger estrogen effects and more of the usual side effects, like breast tenderness, bloating, nausea, headaches, or mood changes.

The important part: this is only theoretical right now. It comes from test-tube research, not from people, and it has never actually been reported in humans. So I wouldn't lose sleep over it, but it's worth mentioning to your pharmacist or doctor before adding a concentrated neem supplement, just so someone keeps an eye on how you feel.
