Lumateperone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Phellodendron interacts with.

Theoretically, phellodendron might increase serum levels of drugs metabolized by CYP3A4.
Phellodendron contains berberine. In vitro research and preliminary clinical research show that berberine moderately inhibits CYP3A4. Theoretically, phellodendron might also inhibit CYP3A4.

Here's the gist with Caplyta (lumateperone): your body clears this medication mainly through a liver enzyme called CYP3A4. The berberine in phellodendron can mildly slow that same enzyme down. So in theory, taking them together could let lumateperone build up a bit more than usual in your bloodstream.

If levels climb, you might notice the drug's common side effects a little more strongly, things like drowsiness, dizziness, dry mouth, or feeling sluggish. I want to be honest, though: this is a theoretical concern based on lab and early research, not confirmed real-world cases, so I wouldn't lose sleep over it. Still, since phellodendron is often combined with other herbs, it's worth a quick chat with your pharmacist or prescriber before adding it. Please don't change your lumateperone on your own.

What Is Lumateperone Used For?

Brand name: Caplyta

Caplyta (lumateperone) is approved for adults with schizophrenia, depressive episodes associated with bipolar I or II disorder (bipolar depression), and as an add-on treatment for major depressive disorder (MDD).

For bipolar depression, Caplyta can be used on its own or alongside lithium or valproate. For MDD, Caplyta is used together with an antidepressant — not on its own as a standalone antidepressant.
