Ashwagandha + Lumateperone
Lumateperone brand name: Caplyta.
Lumateperone falls into 2 drug categories that Ashwagandha can interact with; the most serious is rated moderate. Theoretically, taking ashwagandha might decrease the effects of immunosuppressants.

These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Ashwagandha interacts with whole categories of medication. Lumateperone falls into 2 of those categories, so it can interact with Ashwagandha in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
IMMUNOSUPPRESSANTS
1 of 2Why this applies to Lumateperone
Lumateperone falls under IMMUNOSUPPRESSANTS — the group Ashwagandha interacts with.
Ashwagandha has demonstrated immunostimulant effects in humans. Animal research has shown that ashwagandha can attenuate the immunosuppression caused by cyclophosphamide.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's something worth knowing about this particular pairing. The interaction in our database really speaks to a different group of medicines, the true immunosuppressants (drugs that quiet down your immune system). The concern there is that ashwagandha may gently stimulate immune activity and work against them. Lumateperone (Caplyta) is not an immunosuppressant, though. It's an antipsychotic that works on brain chemistry to help with schizophrenia symptoms, so this specific immune-related concern doesn't really line up with how Caplyta works.
On top of that, the evidence here is theoretical and comes from lab and animal studies, not from people taking these two together. I wouldn't lose sleep over it. Still, ashwagandha can add to drowsiness, and since Caplyta is sedating on its own, that's the more realistic thing to watch. Please run it by your pharmacist or doctor before combining them.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Lumateperone also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that ashwagandha extract induces CYP3A4 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Caplyta (lumateperone): your body relies on an enzyme called CYP3A4 to process this medication. Some early lab research hints that ashwagandha may speed up that enzyme. If that happened in real life, your body could clear lumateperone a little faster, which in theory might make it slightly less effective at controlling schizophrenia symptoms.
I want to be honest about how shaky this is. This concern comes only from test-tube studies, not from people, and it's rated a minor interaction. So I wouldn't lose sleep over it. Still, since lumateperone keeps your symptoms steady, it's worth a quick chat with your prescriber or pharmacist before adding ashwagandha so they can keep an eye on how you're doing.

From our pharmacist’s overview on the Lumateperone page
“Lumateperone (Caplyta) treats schizophrenia and bipolar depression, and because the body clears it mainly through one liver enzyme, CYP3A4, this list deserves real care rather than a quick skim. The three ingredients rated major all hit that vulnerability or the drug itself: grapefruit can push lumateperone levels higher, St.”
