These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Ashwagandha interacts with whole categories of medication. Caffeine (OTC drug) falls into 2 of those categories, so it can interact with Ashwagandha in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Minor).
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
1 of 2Why this applies to Caffeine (OTC drug)
Caffeine (OTC drug) falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ashwagandha interacts with.
In vitro research shows that ashwagandha extract induces CYP1A2 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for these two. Your body clears caffeine mainly through a liver enzyme called CYP1A2. In lab studies, ashwagandha seems to speed that enzyme up a bit. If that holds true in people, your system could break down caffeine a little faster, meaning the pick-me-up from your caffeine pill might wear off sooner or feel weaker than usual.
I want to be honest about the strength of this, though. It's rated minor and comes only from test-tube and animal research, not from real patients, so it may not amount to much day to day. Nobody's going to be harmed here, you just might notice less of a buzz. If you take caffeine for alertness and feel it's not working as well after starting ashwagandha, check in with your pharmacist.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Caffeine (OTC drug) also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that ashwagandha extract induces CYP3A4 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for pairing ashwagandha with a caffeine pill like Vivarin. Lab studies hint that ashwagandha may speed up a liver enzyme called CYP3A4, which can be one of the pathways that helps clear caffeine from your body. If that happens, your caffeine could be broken down a little faster, meaning the pick-me-up (more alertness, less drowsiness) might feel a touch weaker or shorter than usual.
Honestly, I wouldn't lose sleep over this one. The evidence is only from test-tube research, the rating is minor, and caffeine is also handled mostly by a different enzyme (CYP1A2). So any real-world effect is likely small. Still, if you notice your caffeine just isn't doing much, mention it to your pharmacist or doctor.

From our pharmacist’s overview on the Caffeine (OTC drug) page
“The person swallowing a caffeine pill like Vivarin to power through a shift is often the same person drinking coffee, taking an herbal energy blend, or scooping pre-workout powder, and that overlap is exactly where the danger sits. Seven of the 208 supplement ingredients on file carry a major rating, and those seven are worth understan…”
