Lumateperone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the basic idea with Caplyta (lumateperone) and ayahuasca. Your body relies on a liver enzyme called CYP3A4 to break down lumateperone and keep its level in a normal range. The beta-carboline compounds in ayahuasca (like harmine) appear to slow that same enzyme down, at least in lab dish studies. If that happened in a person, lumateperone could build up and hit you harder than usual, meaning more drowsiness, dizziness, dry mouth, or feeling lightheaded when you stand up.

I want to be honest about how shaky this evidence is: it comes from test-tube research, not real people, so it's theoretical. Even so, ayahuasca is potent and risky on its own, and mixing it with an antipsychotic is not something to try alone. Please run it by your doctor or pharmacist first.

Brand name: Caplyta

Caplyta (lumateperone) is approved for adults with schizophrenia, depressive episodes associated with bipolar I or II disorder (bipolar depression), and as an add-on treatment for major depressive disorder (MDD).

For bipolar depression, Caplyta can be used on its own or alongside lithium or valproate. For MDD, Caplyta is used together with an antidepressant — not on its own as a standalone antidepressant.
