Why this applies to Selegiline

Selegiline falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the thing that makes this pair genuinely risky: selegiline is an MAO inhibitor (it blocks MAO-B, and at higher doses or with the Emsam patch it hits MAO-A too). Ayahuasca's vine compounds, harmine and harmaline, are also strong MAO-A inhibitors, and the brew adds DMT, which boosts serotonin activity. Stacking two MAO inhibitors together can let serotonin pile up to dangerous levels.

That raises the risk of serotonin syndrome, with things like tremors, sweating, shivering, a racing heart, confusion, fever, and dangerous spikes in blood pressure. A case of this has been reported with another serotonin drug plus ayahuasca. This is theoretical for selegiline specifically, but the mechanism is real and concerning. Please don't combine these without talking to your doctor or pharmacist first.

Theoretically, ayahuasca might have additive effects when used with dopamine agonists.
Ayahuasca contains the beta-carboline constituent harmine. Animal research shows that harmine increases extracellular dopamine levels.

Here's something worth flagging if you take selegiline for Parkinson's. Selegiline is an MAO-B inhibitor, and ayahuasca's vine ingredient (harmine) is also a monoamine oxidase inhibitor. On top of that, animal studies suggest harmine can push up dopamine levels, and selegiline works by boosting dopamine too. So in theory, the two could stack on top of each other.

The bigger worry is the combined MAOI effect. Mixing MAO inhibitors can spike blood pressure, cause headaches, agitation, rapid heartbeat, or in serious cases a dangerous reaction. The DMT in ayahuasca makes this even riskier. This is based on pharmacology rather than hard human studies, but it's not a combination I'd take lightly. Please talk with your doctor or pharmacist before going anywhere near this.

What Is Selegiline Used For?

Selegiline is used as an add-on treatment for people with Parkinson's disease who are already taking levodopa/carbidopa but are finding that the medication's effects are wearing off or becoming less reliable. It is not used alone — it only works as part of a combination approach.

In clinical studies, adding selegiline helped reduce the amount of daily levodopa needed, cut down on "off" time (periods when symptoms break through), and improved things like tremor, speech, movement, and overall daily function.
