Dapsone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Dapsone: your liver uses an enzyme called CYP3A4 to help break it down. The beta-carboline alkaloids in ayahuasca (harmine and harmaline) can slow that enzyme down, at least in lab studies. If that happened in your body, more dapsone could linger in your blood, which may push up its effects and its side effects.

With dapsone, that could mean more risk of things like methemoglobinemia (a blood oxygen problem causing bluish lips, headache, or shortness of breath) or anemia. I'll be honest though: this is only a theoretical concern seen in test tubes, not in real people. Beyond that, ayahuasca carries its own serious risks. Please loop in your doctor or pharmacist before combining anything.

About Dapsone

What Is Dapsone Used For?

Brand names: Dapsone, Avlosulfon, Aczone

Dapsone tablets are used to treat dermatitis herpetiformis (a chronic, intensely itchy blistering skin condition) and leprosy (all forms, except where proven resistance to dapsone exists). For leprosy, dapsone tablets are typically used in combination with other anti-leprosy medicines.

Aczone Gel (dapsone gel, 7.5%) and dapsone gel are indicated for the topical treatment of acne vulgaris in patients 9 years of age and older. This topical form is applied to the skin and is not used for leprosy or dermatitis herpetiformis.
